WHO的 laboratory methods for SARS diagnosis 不只用於SARS，也是 PCR diagnosis的 golden standards。其中最主要的精神就是要有以下幾個部分的 controls and duplications：

1. RNA extraction 的過程要有 positive and negative controls
2. PCR 的過程要有要有 positive and negative controls **並且加上一個inhibition control (the patient sample spiked with a weak positive control to detect PCR inhibitory substances)
3. duplicate 要從 sample preparation, RNA extraction做起，不可以只在PCR部分做duplicate

如果有以下三種duplicate，可以提高PCR診斷的specificity and reliability

(A)不同實驗室的duplicate
(B)不同部位，不同組織或不同樣本的 duplicate
(C)SARS不同基因片段(不同primers)的 duplicate
